OBJECTIF: Discuter de la pharmacologie, de la pharmacocinétique, et de la place du bortezomib dans le traitement du myélome multiple.

REVUE DE LITTÉRATURE: Une recherche MEDLINE (janvier 1985 à mai 2003) a été effectuée. Des résumés de congrès scientifiques ainsi que des bibliographies d'articles ont également été utilisés pour les fins de cet article. Toutes les données publiées traitant de l'utilisation du bortezomib dans le traitement du myélome multiple ont été considérées. Toutes les études cliniques, particulièrement celles de Phase II, ont été retenues.

RÉSUMÉ: Les traitements actuellement disponibles pour le myélome multiple permettent l'obtention de résultats significatifs mais temporaires. Le bortezomib est un nouvel anti-cancéreux ayant une activité significative dans les cas de myélome multiple en rechute et réfractaires.

CONCLUSIONS: La recherche clinique sur le bortezomib est peu avancée. Toutefois, il représente une nouvelle modalité de traitement pour les patients souffrant d'un myélome multiple et chez qui peu d'alternatives thérapeutiques sont disponibles.
