OBJECTIF: Faire le point sur la pharmacologie, la pharmacocinétique, l'efficacité clinique, et le profil de sécurité de l'aripiprazole dans le traitement de la schizophrénie.

SOURCES DE DONNEES: Information extraite de PubMed (janvier 1995–août 2002). Des résumés, posters, et communications scientifiques ont également été utilisés.

SYNTHESES DES DONNEES: L'aripiprazole est un agoniste partiel de la dopamine et un antagoniste des récepteurs de la sérotonine (5-HT2A), prescrit à la dose de 10 à 30 mg/j, sans phase de titration. Les essais cliniques sur des périodes courtes démontrent une efficacité sur les exacerbations aiguës, et les essais à long terme un maintien des rémissions. La plupart des événements indésirables sont de peu de gravité. L'incidence des symptômes extrapyramidaux est faible, l'akathisie étant le plus courant.

CONCLUSIONS: L'aripiprazole démontre actuellement à la fois efficacité et sécurité dans son utilisation pour la schizophrénie.
