OBJECTIVO: Repasar la farmacología, la farmacocinética, y el lugar en la terapia de bortezomib.

FUENTE DE DATOS: Una búsqueda de la literatura se llevó a cabo en PubMed/MEDLINE para enero 1985 a mayo 2003. También se usaron extractos de reuniones y bibliografías de artículos identificados.

SELECCIÓN DE ESTUDIOS Y EXTRACCIÓN DE DATOS: Toda información publicada relevante a la actividad clínica de bortezomib en mieloma múltiple (MM) fue considerada. Todos los estudios clínicos con énfasis en estudios de Fase II fueron seleccionados.

SÍNTESIS DE DATOS: La terapia actual para MM rinde respuestas significativas aunque temporales. Bortezomib es un agente anticáncer novedoso con actividad significativa en MM en relapso y refractorio.

CONCLUSIONES: Aunque es temprano en los datos de estudios clínicos, bortezomib ofrece una modalidad terapeútica nueva para pacientes con MM que de otra manera tendrían muy pocas opciones.
