DRUG INFORMATION ROUNDS

OBJETIVO: Evaluar la interacción farmacocinética entre tenofovir y didanosina cuando se usa de forma combinada como un régimen de terapia antiretroviral altamente activo (HAART).

FUENTES DE INFORMACIÓN: Se realizo una búsqueda de la literatura a través de MEDLINE entre 1966 a enero 2003 usando términos tales como tenofovir y didanosina. También se repasó extractos y carteles relevantes que se presentaron en conferencias recientes tales como las de la Sociedad Internacional del SIDA, la Conferencia Intercientífica Sobre Agentes Antimicrobianos y Quimioterapia (ICAAC), y la Sociedad Americana de Enfermedades Infecciosas (IDSA).

SÍNTESIS DE INFORMACIÓN: Tenofovir está aprobado para uso en combinación con otros agentes antiretrovirales en el tratamiento de la infección del VIH. La interacción entre tenofovir y didanosina fue evaluada para determinar la seguridad de esta terapia combinada.

CONCLUSIONES: Cuando se usa concurrentemente, tenofovir aumenta la concentración plasmática máxima y el área bajo la curva de concentración contra tiempo de didanosina. Se necesita datos adicionales en pacientes infectados con el VIH para determinar la toxicidad a largo plazo de esta terapia combinada. Se debe de considerar una reducción en la dosis de didanosina cuando se usan estos agentes en combinación.
