PHARMACOKINETICS

OBJETIVO: Presentar un armazón para el diseño y análisis de estudios de farmacocinética de población (PPK) y presentar un ejemplo de la aplicación de un modelo de PPK.

FUENTES DE INFORMACIÓN: Información sobre la farmaocinética de población fue obtenida a partir de MEDLINE (1979–diciembre 2003) y de las bibliografías de artículos de repaso y libros que fueron identificados. Se usó esta información en conjunto con experiencia para explicar los varios enfoques metodológicos usados con PPK.

SÍNTESIS: El diseño y análisis de los estudios de la farmacocinética de población (ie, el desarrollo de modelos de PPK) requiere mucha reflexión. Los modelos son de principalmente 2 tipos: descriptivo y profético. El proceso empleado con estos 2 tipos de modelos es diferente.

CONCLUSIONES: Los modelos de PPK tienen gran utilidad y ofrecen muchas aplicaciones. Son muy distintos a los modelos farmacocinéticos de sujeto único y por lo tanto requieren enfoques diferentes en cuanto a la estimación del modelo.
